Poly(adenosine-diphosphate-ribose) polymerase 1 (PARP1) is a multifunctional nuclear enzyme that plays a crucial role in maintaining genome integrity, responding to cellular stress, and regulating transcription. It is a key player in DNA damage repair, particularly in response to single-strand and double-strand breaks. PARP1's activity is essential for base excision repair, nucleotide excision repair, non-homologous end joining, and homologous recombination repair. Its inhibition is therapeutically exploited in cancers with defective homologous recombination pathways.
PARP inhibitors block PARP1 enzymatic activity, preventing DNA repair, leading to cell death in cells with homologous recombination deficiencies.
