Poly (ADP-ribose) polymerases (PARPs) are a family of enzymes that catalyze the transfer of ADP-ribose units from NAD+ to target proteins, a process known as ADP-ribosylation. This modification plays a crucial role in various cellular processes, most notably DNA repair, chromatin remodeling, transcription regulation, and cell death pathways. PARP1 is the most studied member and is responsible for the majority of cellular poly(ADP-ribosyl)ation activity. PARP inhibitors are used therapeutically, especially where BRCA-mediated double-strand break repair is defective.
Inhibition of PARP enzymatic activity, leading to accumulation of DNA damage and cell death, particularly in cells with defective homologous recombination repair.
