Overview

Polyacrylamide hydrogel is a synthetic, water-swollen polymer network formed by the polymerization of acrylamide with a cross-linking agent like N,N'-methylenebisacrylamide (PubChem, 2024). Within this matrix, water molecules exist in distinct physical states: "bulk water," which retains a tetrahedral hydrogen-bonding network similar to pure liquid water, and "confined water," which is restricted by the polymer chains and exhibits altered vibrational and rotational dynamics (Journal of Physical Chemistry B, 2001). This system is not a biological therapeutic target such as a receptor or enzyme; rather, it is a material used extensively in biotechnology for gel electrophoresis and as a scaffold for tissue engineering (Nature Methods, 2012). In pharmaceutical science, the interaction between the polymer network and the hydrogen-bonding states of water determines the diffusion rates and release profiles of encapsulated drugs (Chemical Reviews, 2012). While the polymerized hydrogel is generally considered non-toxic, the residual acrylamide monomer is a potent neurotoxin and suspected human carcinogen, necessitating rigorous purification for medical applications (NIOSH, 2023). Understanding the behavior of confined water is crucial for optimizing the mechanical stability and biocompatibility of these hydrogels in clinical settings (Journal of Molecular Liquids, 2017).
