The polyadenylation machinery is a multi-protein complex responsible for the addition of a poly(A) tail to the 3' end of eukaryotic pre-messenger RNA (pre-mRNA), a process known as polyadenylation. This modification is essential for producing mature mRNA molecules that are stable, efficiently exported from the nucleus, and competent for translation. The machinery consists of several core complexes including CPSF, CstF, CFIm, and CFIIm, as well as additional factors such as Symplekin, PAP, and PABPN1. It recognizes specific sequence motifs on the pre-mRNA, cleaves the RNA, and adds a poly(A) tail. Alternative polyadenylation, where different poly(A) sites are used, generates transcript diversity and can impact gene regulation. Dysregulation of the polyadenylation machinery has been linked to diseases such as cancer.
