Overview

Polyamine biosynthetic enzymes are responsible for the production of polyamines (putrescine, spermidine, and spermine), which are essential for cell growth, proliferation, and various cellular functions. The core enzymes include ornithine decarboxylase, which catalyzes the first and rate-limiting step, as well as S-adenosylmethionine decarboxylase, spermidine synthase, and spermine synthase. These enzymes are tightly regulated, as polyamines play pleiotropic roles in DNA stabilization, gene expression, apoptosis, and protein synthesis. Dysregulation of polyamine biosynthesis contributes to several diseases, particularly cancers and proliferative disorders, making these enzymes important therapeutic targets. Pharmacological inhibitors of polyamine biosynthetic enzymes, such as DFMO, are being studied and used in clinical contexts to limit pathological cell growth[1][2][4][5][6][7][8].
