Polyamine oxidase (PAO) is a flavin adenine dinucleotide (FAD)-dependent enzyme that catalyzes the oxidation of carbon-nitrogen bonds in secondary amino groups of polyamines. It plays a crucial role in polyamine catabolism and interconversion, maintaining cellular polyamine homeostasis. The enzyme-generated hydrogen peroxide acts as a signaling molecule involved in various biological processes. Targeting PAO is of therapeutic interest, especially in cancer, due to the elevated polyamine uptake and metabolism in cancer cells.
