Polycomb group RING finger protein 2 (PCGF2)

Polycomb group RING finger protein 2 (PCGF2) is a zinc finger-containing transcriptional repressor and an epigenetic regulator encoded by the PCGF2 gene. PCGF2 is a core component of the canonical Polycomb repressive complex 1 (PRC1), which helps mediate chromatin compaction and heritable gene silencing by catalyzing the monoubiquitination of histone H2A at lysine 119. Functionally, PCGF2 plays a critical role in repressing the expression of developmental genes, maintaining stem cell identity, tumor suppression, immune regulation, and endocrine signaling (e.g., regulating progesterone receptor expression to mediate ovulation). Dysregulation or mutation of PCGF2 is implicated in cancers (especially breast cancer, leukemia), reproductive dysfunction, and developmental syndromes such as Turnpenny-Fry Syndrome. PCGF2 interacts with diverse partners and chromatin marks to execute its functions; in some cancers it has been shown to modulate oncogene stability by interfering with post-translational modification pathways. PCGF2 thus represents a key node in epigenetic, developmental, and disease-associated gene regulation[1][2][3][5].
