Overview

Polycomb group RING finger protein 6 (PCGF6) is a member of the Polycomb group family and functions as a key regulator in the PRC1-like complex, which exerts transcriptional repression mainly via chromatin modification, such as monoubiquitination of histone H2A. Unlike canonical PRC1 proteins, PCGF6 binds to promoters with active chromatin marks and has been characterized as a master regulator in maintaining embryonic stem cell identity, positively regulating the expression of core pluripotency factors such as Oct4, Sox2, Nanog, and Lin28. PCGF6’s activity is modulated by cell cycle-dependent phosphorylation, and it interacts with other PcG proteins. Elevated or reduced expression of PCGF6 influences stem cell pluripotency, differentiation, and has roles in various malignancies such as medulloblastoma. There are no specific drugs approved for targeting PCGF6, but its epigenetic regulatory roles render it an attractive target in cancer and regenerative medicine research. Safety concerns center on the potential for wide-reaching effects on gene expression and cell fate due to its central role in epigenetic control.
