Polycystin family receptor for egg jelly (PKDREJ) is a testis-specific protein in humans encoded by the PKDREJ gene. It is a member of the polycystin family, characterized by 11 transmembrane domains, a receptor for egg jelly (REJ) domain, a G-protein-coupled receptor proteolytic site (GPS) domain, and a polycystin-1, lipoxygenase, alpha-toxin (PLAT) domain. PKDREJ is believed to act as a sperm surface receptor for egg zona pellucida glycoproteins, crucial for sperm-egg recognition and regulating the timing of acrosome exocytosis, a process essential for fertilization. While gene paralogs in the polycystin family are implicated in kidney diseases such as polycystic kidney disease, PKDREJ itself is primarily linked to reproductive function, with potential but unproven roles in human fertility and related genetic disorders. There are no known drugs directly targeting PKDREJ, and its therapeutic targeting remains largely unexplored[1][2][4][5][6].
