Overview

The polymerase acidic (PA) protein endonuclease domain is a critical component of the influenza virus RNA-dependent RNA polymerase complex, located at the N-terminus of the PA protein. Its primary function is to cleave host pre-mRNAs to generate capped RNA primers via "cap-snatching," which the viral polymerase then uses to initiate synthesis of viral mRNAs. This activity is essential for both transcription and replication of the influenza virus genome. The PA endonuclease is a validated antiviral drug target.
