Overview

Polymerase delta-interacting protein 3 (POLDIP3) is a multifunctional nuclear protein that serves as an accessory factor in DNA replication and RNA processing. It directly interacts with the DNA polymerase delta complex and proliferating cell nuclear antigen (PCNA) to promote DNA replication, particularly under conditions of replication stress. POLDIP3 is required for the activation and maintenance of DNA damage checkpoints, safeguarding genome integrity and participating in the repair and restart of stalled replication forks. Additionally, the protein contains an RNA recognition motif, facilitating roles in mRNA translation and export, through interaction with ribosomal protein S6 kinase 1 (S6K1) and ALY/REF-like complexes. POLDIP3 is widely expressed and its dysregulation is implicated in cancer development due to its roles in genomic stability and cell cycle control.
