Polypeptide N-acetylgalactosaminyltransferase 11 (GALNT11) is an enzyme that initiates protein O-linked glycosylation by transferring N-acetyl-D-galactosamine to serine or threonine residues of polypeptides, a key step in forming O-glycans[2][3][4]. GALNT11 localizes to the Golgi apparatus and specifically regulates left-right embryonic patterning by mediating glycosylation of the NOTCH1 receptor, which modulates Notch signaling activity[1][2]. This activity influences the balance and identity of motile and immotile cilia at the left-right organizer during development, and disruptions of GALNT11 (by mutation or deficiency) are associated with congenital heart diseases such as heterotaxy and right atrial isomerism[1][2]. GALNT11 acts as a critical developmental regulator and is a member of the polypeptide N-acetylgalactosaminyltransferase family of glycosyltransferase enzymes.
