Overview

Polypeptide N-acetylgalactosaminyltransferase 15 (GALNT15) is an enzyme that catalyzes the initial step in O-linked oligosaccharide biosynthesis by transferring an N-acetyl-D-galactosamine residue from UDP-GalNAc to serine or threonine residues on polypeptide substrates. It shows broader substrate specificity toward mucin peptides, such as Muc1a and Muc5AC, but has lower transferase activity compared to related family members. GALNT15 is part of the large family of polypeptide N-acetylgalactosaminyltransferases that regulate mucin-type O-glycosylation—a modification important in extracellular matrix structure, cell signaling, and protein stability. Mutations or dysregulation may be associated with certain diseases, including classic Ehlers-Danlos syndrome.
