Overview

Polypeptide N-acetylgalactosaminyltransferase 16 (GALNT16) is an enzyme localized primarily to the Golgi apparatus, where it catalyzes the first step of mucin-type O-linked glycosylation by transferring N-acetyl-D-galactosamine residues to serine or threonine residues on protein substrates. GALNT16 is a member of the large GALNT enzyme family, which shows diversity in substrate specificity and tissue expression. Although it has been implicated in cancer and some rare diseases, little is known about its precise substrate specificity, its normal physiological roles beyond glycosylation, or its suitability as a drug target. Currently, there are no drugs targeting GALNT16, but its genetic variants have been investigated for cancer risk association, especially in breast cancer.
