Overview

Polypeptide N-acetylgalactosaminyltransferase like 6 is a 601-amino-acid membrane-bound enzyme expressed mainly in human testis, brain, spinal cord, cerebellum, and skeletal muscle. It initiates O-glycosylation by transferring GalNAc from UDP-GalNAc to serine/threonine residues on specific proteins in the Golgi apparatus, a crucial step in modifying mucins and regulating protein function. This enzyme plays a key role in host-microbiota interactions by facilitating glycan digestion for commensal bacteria, thereby influencing immune modulation, nutrient digestion, pathogen colonization, and energy metabolism. Variants in the GALNTL6 gene (especially rs558129) have been associated with athletic performance traits, particularly in sprint and strength athletes. While its specific disease associations are limited, emerging evidence highlights its importance in metabolic and immune physiology.
