Overview

A polypharmacologic network is not a single molecular entity but rather a complex system of interactions between one or more drugs and multiple biological targets (Hopkins, 2008, Nature Chemical Biology). This approach contrasts with the traditional "one drug, one target" paradigm, acknowledging that many effective medications exert their therapeutic effects by modulating several proteins, enzymes, or receptors simultaneously (Peters, 2013, Journal of Medicinal Chemistry). In the context of complex diseases like cancer or neurodegeneration, polypharmacologic networks are used to map how drugs can overcome robustness and redundancy in biological pathways (PubMed, 2021). By targeting multiple nodes within a network, these therapies can potentially increase efficacy and reduce the likelihood of drug resistance. However, the complexity of these networks also poses significant challenges in predicting off-target effects and managing cumulative toxicities (Reddy & Zhang, 2013, Expert Opinion on Drug Discovery). Ultimately, the study of polypharmacologic networks represents a shift toward systems biology in drug discovery, aiming to optimize the holistic impact of a drug on a disease state.
