Overview

Polyphenol activity refers to the pleiotropic biological effects mediated by polyphenolic compounds, a vast group of plant-derived secondary metabolites characterized by multiple phenolic rings. This term is not a discrete therapeutic target such as a protein or receptor, but rather a description of the chemical and biological properties inherent to this class of molecules (Tsao, 2010). Polyphenols exert their effects through various mechanisms, including the direct scavenging of reactive oxygen species (ROS), the chelation of transition metals, and the modulation of signaling pathways like NF-κB and Nrf2 (Pandey & Rizvi, 2009). They have been extensively studied for their roles in mitigating oxidative stress and inflammation, which are central to the pathogenesis of cancer, cardiovascular disease, and neurodegeneration (NIH NCCIH, 2023). Because of their multi-target nature and lack of specificity, 'Polyphenol activity' is considered an incorrect designation for a drug target in a biotech context. Furthermore, the clinical utility of polyphenols is frequently limited by their poor bioavailability and rapid metabolic transformation within the human body (Teng & Chen, 2019).

Mechanism of action

Polyphenols act by neutralizing reactive oxygen species, chelating pro-oxidant metals, and non-specifically or specifically binding to signaling proteins to modulate pathways like MAPK/ERK, PI3K/Akt, and NF-κB (Tsao, 2010; Pandey & Rizvi, 2009).
