Polypyrimidine tract-binding protein 3 (PTBP3) is an RNA-binding protein of the hnRNP family that binds to CU-rich elements in exons and introns, influencing mRNA splicing, stability, localization, and translation. It is crucial for regulating cell differentiation and post-transcriptional gene expression. PTBP3 is highly expressed in many cancers and is associated with poor prognosis, cancer progression, metastasis, and resistance to chemotherapy. Mechanistically, PTBP3 controls key cancer-related processes such as epithelial–mesenchymal transition, cell migration, and tumor microenvironment interactions. Functionally, PTBP3 can be used as a biomarker for predicting cancer outcomes and may represent a therapeutic target, although intervention carries uncertainties due to its extensive involvement in essential cellular processes.
