Overview

Polysaccharide biosynthesis domain-containing protein 1 (PBDC1), also known as CXorf26 or MGC874, is a conserved human protein encoded by the PBDC1 gene located on chromosome Xq13.3[1][3]. Its precise molecular function in humans remains unclear. The name derives from a conserved DUF757 polysaccharide biosynthesis domain spanning most of the protein, a feature noted in its yeast homolog (YPL225W), but meaningful roles in animal cell polysaccharide synthesis are unproven since humans do not synthesize xylan-like polysaccharides[3]. Instead, evidence suggests PBDC1 participates in cellular processes such as regulation of transcription (notably through interactions with RNA polymerase II), protein ubiquitination, and ribosomal function. These characteristics draw from both human protein-protein interaction data and well-conserved features of the yeast ortholog[1][3][5]. PBDC1 is expressed ubiquitously across human tissues, suggesting a possible housekeeping function. Several protein-protein interactions have been proposed, including with SMAD2, PHB, CTNNB1, BABAM1, BRIX1, and POLR2H proteins, implicating it in signaling, proliferation, and ribosome biogenesis[3]. While the locus neighbors genes linked to X-linked mental retardation syndromes, no direct disease or drug associations are currently established for PBDC1. There is no evidence PBDC1 serves as a therapeutic target, biomarker, or safety concern at this time.
