Polysaccharide ligands are complex carbohydrates that play essential roles in biological recognition, serving as the primary binding partners for a variety of cell-surface receptors including selectins, siglecs, and toll-like receptors (NIH, 2021). These molecules are critical for processes such as leukocyte rolling during inflammation, cell-cell adhesion, and the innate immune response to pathogens (Frontiers, 2023). In medicine, polysaccharide ligands are utilized both as therapeutic agents, such as the anticoagulant heparin, and as targets for vaccine development, where bacterial capsular polysaccharides are used to elicit protective immunity (MDPI, 2022). Additionally, specific glycans like sialyl Lewis X are targeted by antagonists to treat conditions like sickle cell disease and inflammatory disorders (SNMMI, 2019). The interaction between these ligands and their receptors is also exploited for targeted drug delivery, using molecules like hyaluronic acid to deliver cytotoxins to CD44-expressing tumor cells (MDPI, 2023). Despite their therapeutic potential, the structural complexity and widespread distribution of polysaccharides can lead to challenges such as off-target effects, immunogenicity, and complex pharmacokinetic profiles (RSC, 2014).
Polysaccharide ligands function by binding to specific receptors such as selectins, toll-like receptors (TLRs), and C-type lectins to modulate immune signaling and cell adhesion (NIH, 2021). Therapeutic strategies include using polysaccharides as vaccines to induce antibody production, employing them as anticoagulants by activating antithrombin III, or targeting their receptors with monoclonal antibodies and small molecule mimics to inhibit pathological processes like inflammation and metastasis (MDPI, 2022; SNMMI, 2019).
