Overview

The Porcine circovirus type 2 (PCV2) capsid protein, encoded by open reading frame 2 (ORF2), is the sole structural protein of the virus and the primary target for host immune responses. It self-assembles into icosahedral virus-like particles (VLPs) that encapsulate the viral DNA genome and is essential for viral attachment to host cell receptors, such as heparan sulfate, and subsequent entry into the nucleus. PCV2 is the causative agent of porcine circovirus-associated diseases (PCVAD), including postweaning multisystemic wasting syndrome (PMWS), which causes significant economic losses in the swine industry worldwide. Most commercial vaccines utilize recombinant ORF2 protein or inactivated whole virus to elicit neutralizing antibodies and T-cell responses. Continuous genetic evolution of the ORF2 gene has led to the emergence of multiple genotypes (PCV2a through PCV2h), which presents ongoing challenges for long-term vaccine efficacy and diagnostic accuracy.
