Overview

Porcine pancreatic elastase (PPE) is a serine protease enzyme produced in the pancreas of pigs (Sus scrofa), secreted in the intestine, and primarily responsible for the breakdown of elastin and other extracellular matrix proteins. It is a single polypeptide of about 240 amino acids and belongs to the chymotrypsin family of endopeptidases, sharing high sequence and structural homology with other serine proteases like trypsin and chymotrypsin[3][5][7]. PPE cleaves peptide bonds after small, neutral amino acids, with a unique ability to degrade native elastin[2][4]. It is widely used in research as a tool enzyme, particularly for inducing disease models such as emphysema or pancreatitis in rodents, and for screening protease inhibitors, including macrocyclic peptides like FR901451[6]. Despite frequent confusion in the literature, PPE is *not* present in humans and must be distinguished from human pancreatic elastase isoforms[10][1]. It is considered an important biochemical tool and reference serine protease for drug discovery and disease modeling.
