Overview

Porcupine protein (PORCN) is an endoplasmic reticulum-resident O-acyltransferase essential for the palmitoleoylation, secretion, and activity of all mammalian Wnt proteins. It plays a critical role in Wnt signaling pathways, which are involved in embryonic development, tissue renewal, differentiation, and homeostasis. Mutations in the *PORCN* gene are associated with developmental disorders such as Focal Dermal Hypoplasia (FDH)/Goltz-Gorlin Syndrome. PORCN is a therapeutic target, and small-molecule inhibitors are being developed to modulate Wnt signaling in cancer and other diseases.
