Postacrosomal sheath WW domain-binding protein (WBP2NL, also called PAWP) is a testis-specific protein localized to the postacrosomal sheath of spermatozoa, where it plays a key role in oocyte activation during fertilization. WBP2NL shares similarity with WW domain-binding protein 2 (WBP2), but is distinct in its exclusive involvement in reproduction. WBP2NL contains a WW-binding motif, enabling interaction with other proteins, and is essential for the activation of the oocyte immediately after sperm entry, possibly by acting as a sperm factor that triggers calcium oscillations. There is no current evidence supporting WBP2NL as a classic therapeutic or pharmacological target, and it has not been implicated in common disease pathways outside of male infertility. It is not established as a disease biomarker or drug target, and literature does not suggest major challenges, risks, or safety concerns associated with therapeutic targeting of WBP2NL at this time.
