Overview

Potassium channel tetramerization domain-containing protein 12 (KCTD12) is a regulatory protein predominantly expressed during fetal development in tissues such as the brain and cochlea. KCTD12 encodes a protein with a tetramerization (T1) domain, and acts as an auxiliary subunit of GABA(B) receptors in the nervous system, modulating receptor signaling kinetics, G protein coupling (especially βγ subunits), and receptor desensitization. This modulation impacts neuronal excitability, synaptic transmission, and various cognitive functions. KCTD12 is also implicated in cell cycle regulation and tumorigenesis, with emerging evidence supporting its role as a biomarker and therapeutic target in neuropsychiatric disorders (such as bipolar disorder) and various cancers. The protein has a complex role in cellular regulation, bridging neurotransmission and cell proliferation pathways, and participates in multiple protein-protein interactions within receptor complexes.
