Overview

Potassium channel tetramerization domain-containing protein 15 (KCTD15) is a member of the BTB/POZ domain-containing protein family, which forms homo- and hetero-oligomeric complexes (notably with the paralog KCTD1)[2][8]. While sharing homology with proteins that regulate potassium channel assembly, KCTD15 itself is not a canonical ion channel or receptor but rather acts as a regulatory protein impacting diverse biological processes, such as neural crest cell development, negative regulation of specific transcription factors (notably AP-2), and modulation of the NF-κB signaling pathway[3][4][5][9]. Pathogenic variants in the KCTD15 gene are implicated in congenital disorders like aplasia cutis congenita and cardiac malformations, and changes in KCTD15 expression are associated with cancer and metabolic conditions such as obesity and diabetes[2][4][5]. The protein is broadly expressed during development and has roles in formation of skin appendages, cranial structures, and cardiac anatomy, reflecting its critical regulatory function in embryogenesis and tissue differentiation[2][3][5]. There are currently no known drugs targeting KCTD15, and it is not a classical therapeutic target as defined by receptor, enzyme, or ion channel status.
