Potassium channel tetramerization domain-containing protein 20 (KCTD20) is a member of the KCTD family, characterized by a conserved N-terminal BTB (Bric-à-brac, Tram-track, Broad complex) domain that mediates protein-protein interactions and often functions as an adaptor for Cullin3-based E3 ubiquitin ligase complexes[2][3]. Unlike classical ion channels, KCTD20 does not constitute a potassium channel but shares homology with the tetramerization domain (T1) found in voltage-gated potassium channels, which determines oligomerization specificity[1][2]. KCTD20 is predicted to promote phosphorylation of AKT family members and is involved in cytoplasmic signaling processes, with localization to the cytoplasm and association with the cell membrane[3]. The protein is implicated in the pathogenesis of certain neurodegenerative diseases, such as progressive myoclonus epilepsy, but its precise biological roles and potential as a drug target are largely uncharacterized[3].
