Potassium channel tetramerization domain-containing protein 21 (KCTD21) is a member of the KCTD family, defined by the presence of a BTB/POZ domain at the N-terminus, which is involved in protein-protein interactions and facilitates the formation of multiprotein complexes. KCTD21 is thought to act as a substrate-specific adaptor for a BTB-CUL3-RBX1 E3 ubiquitin ligase complex, mediating the ubiquitination and subsequent proteasomal degradation of specific target proteins, including histone deacetylase 1 (HDAC1). The KCTD21 gene is primarily involved in processes such as the negative regulation of the Smoothened signaling pathway and ubiquitin-dependent protein catabolism. While many KCTD family proteins, including closely related KCTD11, have been implicated in the regulation of signaling pathways relevant to cancer and other diseases, the precise pathological significance and therapeutic potential of KCTD21 itself remain under investigation. There are currently no documented, clinically approved drugs targeting KCTD21, nor well-established safety concerns or biomarker roles described in the literature.
