The potassium-chloride cotransporter 1 (KCC1) is a membrane protein predominantly expressed in erythrocytes and other cells, responsible for the electroneutral symport of K^+ and Cl^- ions across the plasma membrane. KCC1 plays a central role in regulating cell volume, maintaining ionic balance, and ensuring proper intracellular chloride concentrations. In red blood cells, its overactivity contributes to cell dehydration and sickling in sickle cell disease. KCC1 belongs to the cation-chloride cotransporter family (SLC12), with isoforms KCC1–4 sharing structural features. KCC transport is tightly regulated by phosphorylation and cellular conditions such as swelling, acidification, and oxygenation state[1][2][4][6]. KCC1 function is implicated in the pathophysiology of hematological disorders and tumor progression, making it a potential but not yet clinically targeted therapeutic protein[1][4].
