Kir6.1 is a pore-forming subunit of ATP-sensitive potassium (KATP) channels, which are crucial in linking cellular metabolism to membrane excitability. These channels are octameric complexes composed of four Kir6.x subunits (either Kir6.1 or Kir6.2) and four regulatory sulfonylurea receptor (SUR) subunits. Kir6.1 is predominantly expressed in vascular smooth muscle, where it forms functional KATP channels with SUR2B regulatory subunits, regulating vascular tone by controlling membrane potential in response to metabolic changes. Mutations affecting either KCNJ8 or its partner genes can lead to diseases such as Cantú syndrome.
