Overview

Potassium voltage-gated channel interacting protein 4 (KCNIP4) is a small calcium-binding protein that functions as an auxiliary subunit for voltage-gated A-type potassium (Kv4) channels, especially prevalent in brain and heart tissues. KCNIP4 contains EF-hand domains characteristic of calcium sensor proteins, and directly regulates Kv4 channel kinetics (activation, inactivation, and recovery) and trafficking to the cell membrane in response to intracellular calcium levels. These interactions have significant effects on neuronal excitability and signal integration. KCNIP4 also binds to presenilin proteins—implicated in neurodegenerative diseases—and is expressed in select neuronal networks. Genetic variants in KCNIP4 have been linked to psychiatric disease susceptibility, antidepressant side effects, and asthma risk. No drugs are known to target KCNIP4 directly, but its regulatory influence over Kv4 channels positions it as a potential candidate for modulating excitability disorders and as a biomarker for certain neuropsychiatric and airway disorders.
