Potassium voltage-gated channel subfamily J member 11 (KCNJ11) encodes the Kir6.2 protein, a key component of ATP-sensitive potassium (KATP) channels. These channels are integral membrane proteins that allow potassium ions to flow more readily into the cell than out. The activity of these channels is regulated by G-proteins and they function in association with sulfonylurea receptors (SUR), forming hetero-octameric complexes critical for cellular excitability and insulin secretion. Mutations or polymorphisms in KCNJ11 are linked with congenital hyperinsulinism, permanent neonatal diabetes mellitus and type II diabetes mellitus.
Sulfonylureas bind to the SUR subunit of the KATP channel, inhibiting its activity and promoting insulin secretion.
