POTE ankyrin domain family member A (POTEA) is a primate-specific protein-coding gene located on chromosome 8p11.1, belonging to the POTE gene family with 14 human paralogs[1][4]. POTE proteins are characterized by an N-terminal cysteine-rich region, multiple ankyrin repeats (protein-protein interaction modules), and a C-terminal spectrin-like domain, suggesting roles in protein-protein and membrane interactions[1][3]. POTEA is the closest human relative to the ancestral ANKRD26 gene, uniquely retaining certain ancestral exons and genomic features[1]. POTEA and other POTE family members show predominant expression in reproductive tissues (prostate, ovary, testis, placenta) and are overexpressed in various cancers[1]. Their precise biological function remains unclear, though some family members have been implicated in pro-apoptotic signaling mediated by cytoskeletal interactions[1]. POTEA is not classically described as a therapeutic receptor, enzyme, transporter, or transcription factor, and currently has no known drug interactions or defined mechanism of action, but may be investigated for its biomarker potential in oncology[1][2][4].
