Overview

POTEE (POTE ankyrin domain family member E) is a member of the human POTE gene family, which encodes primate-specific, cancer/testis antigens characterized by ankyrin repeats and spectrin-like helices[1][3][4]. POTEE is predicted to bind protein kinases and act as a structural component of the postsynaptic actin cytoskeleton[1][3][5]. Its physiological expression is highly restricted to embryonic stem cells and select adult tissues (testis, ovary), but it is aberrantly upregulated in various cancers (notably colorectal, ovarian, pancreatic, lung, and breast), where it functions as an oncogene promoting cell proliferation, migration, invasion, cell-cycle progression, and resistance to apoptosis, through activation of signaling pathways including SPHK1/p65, PI3K/Akt/GSK-3β/β-catenin, and S1P/NF-κB[1][2]. POTEE is under evaluation as a diagnostic and prognostic biomarker and a potential therapeutic target in oncology due to its cancer-specific expression and role in disease progression, but no approved targeted therapies or specific clinical safety data currently exist[1][2].
