Overview

POTE ankyrin domain family member F (POTEF) is a primate-specific member of the POTE ankyrin domain-containing gene family. Its protein product is a chimeric molecule that includes ankyrin repeat domains and an acquired β-actin domain due to retrotransposition, resulting in high homology with the related protein POTEE[3][5]. POTEF is predicted to function in protein kinase binding and as a structural constituent of the postsynaptic actin cytoskeleton, with roles in axonogenesis and cell motility[1][2][6]. Its expression is found in tissues such as ovary, testis, and many cancers, and it localizes to the cytoskeleton near the inner plasma membrane[1][3]. POTEF has been shown to interact with the chaperonin containing TCP-1 complex (CCT), impacting cell proliferation and possibly contributing to cell death through a mitochondrial-dependent apoptotic pathway, especially when overexpressed[1][3]. Overaccumulation may result in autophagic failure in granulosa cells, which is linked to follicular atresia and primary ovarian insufficiency[3]. POTEF is not a classical therapeutic target such as a receptor, enzyme, or transporter, but it is of interest due to its roles in apoptosis, cell proliferation, and as a potential autoantigen in ovarian autoimmunity and some cancers[3][1][2]. There are currently no known drugs targeting POTEF, and it is not established as a clinically actionable biomarker or therapeutic target at this time.
