POTE ankyrin domain family member G (POTEG) is a member of the human POTE gene family, which encodes proteins predominantly expressed in reproductive tissues such as prostate, ovary, testis, and placenta, as well as in various cancers[1][2][3]. POTEG features several ankyrin repeats and a C-terminal spectrin-like domain, and is thought to be involved in protein-protein and cell membrane interactions[3]. Functional studies indicate that POTEG acts as a pro-apoptotic factor: downregulation of POTEG results in decreased apoptosis and poorer prognosis in esophageal squamous cell carcinoma (ESCC)[1]. It appears to suppress tumorigenesis by arresting the cell cycle at the G1/S checkpoint, downregulating cyclin-dependent kinases (CDKs), and inhibiting metastasis by reducing epithelial-mesenchymal transition[1]. POTEG and other POTE family members may serve as biomarkers for disease progression in certain cancers, but there is no current evidence to suggest that POTEG itself is a therapeutic target, nor are there known drugs directly targeting it[1][2][3].
