Overview

POTE ankyrin domain family member H (POTEH) is a member of the primate-specific POTE gene family encoding cancer/testis antigens, characterized by the presence of ankyrin repeats and spectrin-like alpha helices[4][6][3]. These multidomain proteins are primarily expressed in testis, ovary, prostate, and placenta, but are aberrantly overexpressed in several types of cancer, where they are associated with disease progression and poor prognosis[4][6]. POTEH—and related family members—function mainly as scaffold or adaptor proteins; they are implicated in apoptosis, with increased expression observed in apoptotic germ cells and tumor cells[3][4]. Expression profiles and structural features suggest roles in cytoskeletal organization and protein-protein interactions within the tumor microenvironment[1][6]. As a cancer/testis antigen, POTEH is a potential diagnostic and therapeutic target, including for immunotherapy approaches in oncology[6]. However, functional redundancy among family members and some expression in normal tissues present therapeutic challenges and safety concerns[6][4].
