Overview

POU2F2 is a pivotal transcription factor in lymphoid cells, especially B cells, where it is essential for immunoglobulin gene transcription and B-cell differentiation. It binds to the octamer sequence 5’-ATTTGCAT-3’ present in immunoglobulin gene promoters. Through activation of transcription, including anti-apoptotic genes, it sustains immune cell survival and maturation. POU2F2 plays a major role in various cancers, where its dysregulation leads to altered differentiation and cell survival. Furthermore, POU2F2 modulates cytokine gene expression and can affect immune checkpoints in malignancies. In viral infections like EBV, POU2F2 facilitates maintenance of latency through chromatin modification and enhancer binding. Outside immune tissues, its abnormal expression influences proliferation and metabolic reprogramming in diverse cancers and may regulate neuronal differentiation.
