Overview

POU2F2 antisense RNAs (POU2F2-AS1, POU2F2-AS2) are long non-coding RNAs transcribed on the opposite DNA strand to the POU2F2 gene, a key B-cell transcription factor. These lncRNAs regulate gene expression at multiple levels, including epigenetic silencing through DNA methylation, manipulating chromatin architecture via enhancer-promoter looping, and possibly participating in ceRNA networks to sponge microRNAs. Their dysregulation has been implicated in cancer progression: they influence cell proliferation, invasion, and migration, and high expression often correlates with poor prognosis. Although not classical receptors or enzymes, their vital regulatory roles make them attractive experimental therapeutic targets in oncology. No direct clinical drugs currently target these molecules, but antisense oligonucleotides and related RNA-modifying strategies have shown promise in preclinical studies.

Mechanism of action

Gene expression inhibition by antisense interaction. Chromatin remodeling through recruitment of DNMT1 (DNA methyltransferase 1) for epigenetic silencing. Disruption of enhancer-promoter looping, thereby modulating neighboring gene expression. Potential miRNA sponging effect in ceRNA networks (widely reported for lncRNAs, not confirmed for these specific antisense RNAs).
