Overview

POU class 4 homeobox 1 (POU4F1), also known as BRN3A, is a neural-specific transcription factor belonging to the POU-IV family of homeobox proteins[1][3][5]. It is crucial for the development, differentiation, and survival of certain sensory and central nervous system neurons, particularly retinal ganglion cells and sensory neurons in the peripheral nervous system[1][5]. POU4F1 regulates cell fate and survival via transcriptional activation or repression of downstream genes associated with apoptosis (e.g., BCL2), neuronal differentiation, and axon projection[1][3][5]. Dysregulation of POU4F1 is implicated in several cancers, including melanoma—where it confers resistance to BRAF inhibitors through upregulation of the MEK/ERK pathway[2][3][4]—as well as leukemia, breast, cervical, prostate, Ewing’s sarcoma, and thyroid cancers[3].\n\nBeyond its oncogenic roles, germline mutations in POU4F1 are associated with rare neurological syndromes involving ataxia and cerebellar dysfunction[1][3]. As a biomarker, POU4F1 expression and methylation status can mark tumor progression and therapy resistance in various malignancies[2][3].
