Overview

POU class 4 homeobox 2 (POU4F2), also known as BRN3B, is a POU-domain transcription factor critically involved in the regulation of gene expression in specific neural and sensory tissues. Its key biological role is in the differentiation and survival of retinal ganglion cells, and it binds to DNA at specific octamer motifs to control transcriptional activity, acting as either an activator or repressor depending on context[1][2]. POU4F2 also plays a role in cardiac development and response to stress, particularly in the fetal and injured heart, and is re-expressed during cardiac stress responses[3]. It interacts with transcription factors such as p53 and Estrogen receptor alpha, influencing processes like apoptosis and hormonal regulation[1][2]. Aberrant expression of POU4F2 has been linked to accelerated tumor growth in breast cancer and to specific neurological and visual disorders[1][2][3]. The protein has not been directly targeted by therapeutics as of current knowledge, but its expression pattern can serve as a biomarker in certain cancers and developmental disorders[1].
