POU6F2 antisense RNA 2 (POU6F2-AS2)

Target
POU6F2-AS2
Molecular classification
Long noncoding RNA (lncRNA), Antisense RNA
01

Overview

POU6F2 antisense RNA 2 (POU6F2-AS2) is a cytoplasmic long noncoding RNA expressed at significantly higher levels in NSCLC tumors compared to adjacent normal tissue[1][2]. Functionally, it acts as a *competitive endogenous RNA (ceRNA)*, sponging miR-125b-5p, which leads to de-repression and increased expression of E2F transcription factor 3 (E2F3), a driver of cell proliferation, colony formation, and tumor cell motility[1][2]. Knockdown of POU6F2-AS2 inhibits these cancer-promoting processes. Its high expression is correlated with more aggressive disease and poor prognosis, suggesting utility as both a disease biomarker and a candidate therapeutic target in lung cancer[1][2].

Mechanism of action

Not applicable for approved drugs. Mechanistically, antisense oligonucleotides, RNA interference, or targeted lncRNA inhibitors *could* theoretically be used to inhibit its function, but these are preclinical strategies and not validated therapies.

03

Biological functions

Acts as a microRNA sponge (miR-125b-5p)Regulates cellular proliferationModulates cell migration and invasionInfluences oncogenic transcription factor expression (notably E2F3)
04

Disease associations

Cancer (specifically, non-small-cell lung cancer—NSCLC; also upregulated in lung adenocarcinoma and squamous cell carcinoma)Potential role in other malignancies as supported by lncRNA family roles, but direct evidence focused on lung cancer
05

Safety considerations

Not definedpotential challenges include specificity of lncRNA-targeted therapies and off-target effects, as is common for lncRNAs
06

Biomarkers

POU6F2-AS2 expression itself is a potential biomarker for NSCLC diagnosis and prognosisalso may predict aggressive phenotypes
