PP2A Aalpha and B55A interacting phosphatase regulator 1 (PABIR1) is a protein that acts as an inhibitor of serine/threonine-protein phosphatase 2A (PP2A). It inhibits PP2A activity by blocking the substrate binding site on the regulatory subunit PPP2R2A and the active site of the catalytic subunit PPP2CA. Through this inhibition, PABIR1 modulates cell cycle progression, particularly the mitotic G2/M transition checkpoint, by promoting ubiquitin-mediated proteolysis of specific cell cycle regulators such as WEE1. It positively regulates cell growth and the proteasomal degradation of PP2A catalytic subunits, thereby influencing diverse processes related to cell proliferation and division[1][6][7].
