Overview

PPFIB scaffold protein 2 (PPFIBP2), also known as liprin-beta-2, is a member of the liprin family of scaffold proteins that participate in organizing multiprotein complexes, particularly at synaptic sites and focal adhesions[6][5][9]. PPFIBP2 acts as a scaffold in neuronal development, playing a central role in the recruitment and spatial organization of receptor-type protein tyrosine phosphatases, such as LAR and its relatives, to the plasma membrane, thereby influencing axon guidance and synapse formation[5][6][9]. In addition to its developmental functions, PPFIBP2 has been implicated in disease processes: it acts as a tumor suppressor in some cancer types and has been identified as a fusion partner with several oncogenic kinases and phosphatases, suggesting potential relevance to cancer progression and prognosis[4]. While the protein is not a typical direct therapeutic target such as a receptor or enzyme, alterations in its expression or genetic status may have clinical utility as a biomarker in oncology[4][6]. No drugs specifically target PPFIBP2, nor are there established mechanisms of action or reported safety concerns associated with its modulation.
