PPM1K divergent transcript (PPM1K-DT) is a long non-coding RNA transcribed from a region divergent to the protein-coding PPM1K gene[1][3][5]. As a lncRNA, PPM1K-DT is not known to encode a functional protein but may regulate local gene expression, including that of the adjacent PPM1K gene, which encodes a mitochondrial phosphatase crucial for BCAA metabolism. Expression and methylation of PPM1K-DT are altered in several cancer types, particularly prostate cancer, and are associated with epigenetic regulation of disease prognosis, suggesting a functional role in oncogenic or tumor suppressor pathways via modulation of gene regulatory networks[6]. The transcript's biological and clinical significance beyond these observations remains to be comprehensively elucidated, and no drugs or direct molecular interactions are currently described.
