PPP2R2B antisense RNA 1 (PPP2R2B-AS1) is a long non-coding natural antisense transcript bidirectionally transcribed at the PPP2R2B locus, overlapping with exon 7 of the PPP2R2B protein coding gene. The expanded form of this RNA, containing pathological CUG repeats, is associated with neurotoxicity and may contribute to the pathogenesis of spinocerebellar ataxia type 12 (SCA12). Expanded PPP2R2B-AS1 forms RNA foci that sequester splicing factors, induces apoptosis in neuronal models via altered RNA-protein interactions, and can produce proteins via repeat-associated non-ATG (RAN) translation. The transcript is polyadenylated, and full-length characterization is ongoing. Therapeutic targeting of PPP2R2B-AS1 may represent a potential strategy in SCA12, but its low abundance and incomplete annotation present experimental challenges.
