PR domain zinc finger protein 14 (PRDM14) is a key transcription factor and epigenetic regulator primarily expressed during early embryonic development and in primordial germ cells (UniProt: Q9GZV8). It is essential for maintaining pluripotency and driving the epigenetic reprogramming required for germline specification, largely through the regulation of DNA methyltransferases and histone modification (PubMed: 18524774). In adult tissues, PRDM14 expression is generally silenced, but its aberrant reactivation is observed in several high-grade malignancies, including breast, pancreatic, and lung cancers, where it acts as an oncogene by promoting cell survival and metastasis (PubMed: 22869144). Because PRDM14 is a transcription factor—a class of proteins traditionally considered undruggable by small molecules—therapeutic strategies have shifted toward targeting its mRNA. Experimental treatments using small interfering RNA (siRNA) delivered via lipid nanoparticles have shown promise in preclinical models by inducing mRNA degradation, thereby reducing PRDM14 protein levels and inhibiting tumor growth (PubMed: 28341771). These RNAi-based approaches aim to suppress the oncogenic signaling pathways driven by PRDM14, potentially offering a novel treatment modality for refractory cancers.
