Overview

PRDM2 (PR domain zinc finger protein 2) is a gene that produces two main protein products, RIZ1 and RIZ2, through the use of alternative promoters (UniProt Q13029). While RIZ1 contains a PR (SET) domain with histone methyltransferase activity and functions as a tumor suppressor, RIZ2 lacks this domain and is frequently overexpressed in various malignancies, where it acts as a pro-proliferative factor (PubMed 10866308). The RIZ2 isoform mRNA is considered a therapeutic target because its selective knockdown can potentially restore the balance between the tumor-suppressive RIZ1 and the oncogenic RIZ2 (PubMed 25600165). Research indicates that an imbalance in the RIZ1/RIZ2 ratio is a common feature in cancers such as breast, liver, and colon cancer (PubMed 15108055). Therapeutic strategies targeting RIZ2 mRNA typically involve the use of antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs) to specifically degrade the RIZ2 transcript. By reducing RIZ2 levels, these interventions aim to inhibit tumor growth and sensitize cancer cells to other treatments.
