Overview

PRDM2 (PR domain zinc finger protein 2) is a gene that encodes two distinct protein isoforms, RIZ1 and RIZ2, through the use of alternative promoters (UniProt Q13029). RIZ1 contains a PR domain with histone H3 lysine 9 (H3K9) methyltransferase activity and is widely recognized as a tumor suppressor (He et al., 1998, PubMed: 9790530). Conversely, the RIZ2 isoform lacks this PR domain and functions as an oncogene, promoting cell proliferation and survival in various human cancers, including breast, liver, and colon malignancies (Kim et al., 2015, PubMed: 25600165). The mRNA encoding the RIZ2 isoform is a specific target for therapeutic intervention using RNA-based technologies like antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs). These modalities are designed to selectively degrade the RIZ2 transcript or block its translation, thereby restoring the balance between the tumor-suppressive RIZ1 and oncogenic RIZ2 (Pastural et al., 2007, PubMed: 17332325). This approach aims to inhibit tumor growth while minimizing the risk of silencing the beneficial RIZ1 isoform, which shares significant sequence homology with RIZ2 except at the 5' terminus.
