Overview

The PRAME peptide–MHC complex is a cell surface complex resulting from the intracellular processing of the PRAME protein and the subsequent presentation of its derived peptides (notably ALYVDSLFFL and others) by MHC class I molecules on the surface of tumor cells. This allows recognition and targeting by cytotoxic T lymphocytes (CTLs) specific for the complex and by engineered T cell receptors or TCR mimic antibodies. Because PRAME is expressed at high levels in various cancers but only at low levels in normal adult tissues, the PRAME peptide–MHC complex is considered a promising and specific target for cancer immunotherapy, including the development of TCR-based therapies and antibody therapeutics[3][4][6][8][9]. The specificity and immunogenicity are determined by both the peptide sequence and the presenting HLA allele (most often HLA-A*02:01); thus, patient selection considers PRAME expression and HLA type. Therapies targeting this complex must balance efficacy with potential cross-reactivity and immune-related side effects.
